Short description of research interest:
I have been achieved research experiences in receptor signalling and research using cell culture system is my basic experience. In order to contribute towards the success of my potential, I desire to obtain a post doctoral experience in biomedical research and Image analysis which further can complement my research career. Besides research, I want to work in clinical practices.
Since December 2007, I have started a pioneering research to investigate molecular mechanisms of exotoxin A action in human ovarian cancer. Bacterial products such as Exotoxin A have been used in an effort to reduce the growth rate of tumors as because it can be able to trigger apoptosis. However, the mechanism by which ETA induce apoptosis has not yet emerged. In this project, I aim to investigate cytotoxic effects at cellular level using different ovarian cell lines and to reveal the cell signal transduction pathway involved behind exotoxin A induced apoptosis. Further in continuation, I am analyzing PRL signalling pathway in ovarian cancer cell line as because I found that PRL signaling can induce the expression of c-Myc in the cell line treated with exotoxin A.
In November 2007, I defended my thesis entitled ‘’Exoenzyme of Pseudomonas aeruginosa: cellular target s and interaction with 14-3-3’’. My research interest was to analyze how ExoS induce cytotoxicity in host using structural and biochemical observation on survival and death signal transduction pathways e g RAS signalling, AKT signalling and apoptosis signalling. Cell lines were used in order to understand the functional and mechanical activation of ExoS-ADP- ribosyltransferase activity and cytotoxicity at the cellular level and to reveal critical residues of ExoS responsible for cell cytotoxicity and to elucidate the critical signalling targets of ExoS by which it mediates cell death. The research was performed in national as well as international collaborations and studies also extended to the pathogenesis experiment in mouse model and structural analysis of ExoS/14-3-3 molecules. The structural analysis of ExoS/14-3-3 molecules provide information to develop therapeutic against Pseudomonas aeruginosa infection and this study was performed with collaboration in Dortmund Germany. Further, the ExoS induced pathogenesis study was investigated in more relevant clinical model such as in a mouse with acute pneumonia model, performed with collaboration in Northwestern University, Chicago, USA which provide supportive evidences for the cytotoxicity analysis at cellular level and structural analysis.
